Preparation of the selenomethionine derivative of tabtoxin resistance protein

In order to obtain phase information for the X-ray diffraction of tabtoxin resistance protein (TTR) crystal using the MAD phasing method, a selenomethionine (Se-Met) derivative of TTR was overexpressed in E. coli strain M15, with pQE-30 plasmid, through IPTG induction in M9 medium containing Se-Met. The product was purified to an estimated homogeneity of greater than 95% according to SDS-PAGE, by a Ni-NTA metal affinity followed by a Mono Q anion exchange column chromatography. The successful substitution of Se-Met for methionine (Met) was confirmed by MALDI-TOF and ESI-Quadrupole Mass Spectrometry analysis. The derivative crystal was obtained using similar conditions as those for the native.     

The cytogenetics and fiber properties of an allohexaploid betweenGossypium hirsutum andG. somalense

Cytogenetics and fiber properties were studied on the Gossypium hirsutum×G. somalense F₂ hybrid and later generations. The cytological analyses of pollen mother cells (PMCs) were made at meiotic metaphase Ⅰ. The results indicated that the hybrid was a new allohexaploid, its chromosome number determined as 2n=6x=78, and genome group as 2[(AD)₁E₂]. Chromosomal configurations of the hexaploid averaged 0.15Ⅰ+ 38.72Ⅱ+ 0.11Ⅲ + 0.02Ⅳ per cell, 85.09% PMCs having 39 bivalents, and only 11.84% PMCs having 1 to 2 multivalents, indicating that heterogenetic recombinations of chromosomes occurs between (AD)₁ and E₂, nevertheless, the frequency is lower. The hexaploids are self-fertile and the progenies remain the hexaploids, whose fibers are light brown and have higher strength by measurement of HVI 900 system. The fiber strength increases 42% than that of upland cotton variety. Therefore, it is possible for the hexaploid to be an important germplasm resource to improve fiber strength of cotton.     

Expression, purification of a synthetic fuse-protein-TSF from PF4 and TSP1 fragments and its effect on angiogenesis and tumor growth

Sequences encoding PF4 (58–70) and TSP1 (429–459) were linked to yield a single gene TSF which encodes the fuse-protein of TSF. The gene was cloned into a pGEX-2T expression vector to generate a protein GST-TSF, which was strongly expressed inE. coli. The purified GST-TSF was degraded with thrombin to generate the protein TSF. With the methods of MTT and wound repair assay, the effects of TSF on the proliferation and migration of EC were detected, respectively. The results showed that TSF significantly suppressed BAEC proliferation and migration in a dose-dependent manner. The fuse protein GST-TSF, and the peptides PF4 (58–70) and TSP1 (429–459) also inhibited BAEC proliferation and migration, respectively, but their inhibition rates were not as high as TSF. Using the CAM assay, it was shown that TSF, GST-TSF, PF4 (58–70) and TSP1 (429–459) inhibited angiogenesis in chick CAM potentially, the effect of TSF was the highest.In vivo, the growth of Lewis lung carcinoma was potently inhibited by TSF treatment, and the inhibition rate was 68.75% at a dose of 1.00 μmol/kg · d. These findings suggest that the design on TSF gene was successful, and TSF with its anti-angiogenic and anti-tumor activity, should be a useful source of the inhibitor.     

Isolation, purification and characterization of secondary structure of antifreeze protein fromAmmopiptanthus mongolicus

Antifreeze protein (AFP) from Ammopiptanthus mongolicus leaves was isolated and purified with DEAE cellulose 52, Sephacryl 300, heat treatment and preparative isotachophoresis. The molecular weight of AFP was 50 ku by SDS-PAGE measurement. The average thermal hysteresis activity (THA) of AFP was 0.35℃ at 5 mg/mL using a differential scanning calorimeter (DSC). This AFP showed heated stability. From circular dichroism (CD) of AFP spectral data from 195—240 nm, a well-defined secondary structure of 11% a-helix, 34% antiparallel b-sheet and 55% random coil for the plant AFP was deduced.     

Preparation and optical properties of composite thin films with embedded InP nanoparticles

InP nanoparticles embedded in SiO₂ thin films were prepared by radio-frequency magnetron co-sputtering. We analyzed the structure and growth behavior of the composite films under different preparation conditions. X-ray diffraction and Raman spectroscopy analyses indicate that InP nanoparticles have a polycrystalline structure. The average size of InP nanoparticles is in the range of 3–10 nm. The broadening and red shift of the Raman peaks were observed, which can be interpreted by the phonon confinement model. Optical transmission spectra indicate that the optical absorption edges of the films can be modulated in the visible light range. The marked blue shift of the absorption edge with respect to that of bulk InP is explained by the quantum confinement effect. The theoretical values of the blue shift predicted by the effective mass approximation model are different from the experimental results for the InP-SiO₂ system. Analyses indicate that the exciton effective mass of the InP nanoparticles is not constant and is inverse relative to the particles radius, which may be the main reason that results in the discrepancy between the theoretical and the experimental result. We discussed the possible transition of the direct band gap to the indirect band gap for InP nanoparticles embedded in SiO₂ thin films.

岩石材料的非均质性与动态参数

对四种岩石进行了超声波测试。从同一岩块加工的试样尽管宏观上没有明显差异,但超声波通过时间的差异最大可以达到9%将岩样间具有相同性质的长度相抵后,可以认为时间差异只是发生在局部材料,即岩样是由力学特性差异极大的材料构成的。而岩样的动态、静态参数是以不同的方式反映岩样内部材料的力学性质,因而并不会相等。由超声波速度计算的泊松比变动范围较大(最大43%),难以运用;动态模量总是大于变形模量,而与弹性模量的差异可以达到±15%以上。
